چکیده
BackgroundPanton–Valentine leucocidin (PVL)-positive community-associated meticillin-resistant Staphylococcus aureus (CA-MRSA) is increasingly associated with infection outbreaks.AimTo investigate multiple suspected PVL-positive CA-MRSA outbreaks using whole-genome sequencing (WGS).MethodsForty-six outbreak-associated isolates from 36 individuals at three separate Irish hospitals (H1–H3) and incidents involving families H2 were investigated by multi-locus sequence typing (wgMLST).FindingsTwo clusters (CH1 CH2) consisting of 8/10 6/6 t008 ST8-MRSA-IVa H1 H2, respectively, identified. Within each cluster, neighbouring separated ?5 allelic differences; however, ?73 differences identified between the clusters, indicating two independent outbreaks. Isolates H3 maternity unit formed (CH3–SCI CH3–SCII) composed four PVL-negative t4667 ST5-MRSA-V 14 t002 ST5-MRSA-IVc isolates, respectively. ?24 differences, whereas both 1822 distinct Eight t127 ST1-MRSA-V+fus t267 ST97-MRSA-V+fus H2-associated FC1 (N = 4) FC2 7) (FC1 (t127), (t127) (t267)). Neighbouring within closely related exhibited ?7 differences. Intrafamilial transmission was apparent, but detection ?48 indicated no interfamilial transmission.ConclusionThe frequent importation into healthcare settings, association a serious ongoing concern. Community-associated meticillin-resistant Staphylococcus aureus.
Meticillin-resistant Staphylococcus aureus (MRSA) is endemic in hospitals worldwide, and causes substantial morbidity and mortality. Health-care-associated MRSA infections arise in individuals with predisposing risk factors, such as surgery or presence of an indwelling medical device. By contrast, many community-associated MRSA (CA-MRSA) infections arise in otherwise healthy individuals who do ...
متن کاملWhole-genome sequencing for analysis of an outbreak of meticillin-resistant Staphylococcus aureus: a descriptive study
BACKGROUND The emergence of meticillin-resistant Staphylococcus aureus (MRSA) that can persist in the community and replace existing hospital-adapted lineages of MRSA means that it is necessary to understand transmission dynamics in terms of hospitals and the community as one entity. We assessed the use of whole-genome sequencing to enhance detection of MRSA transmission between these settings....
متن کاملCommunity-acquired meticillin-resistant Staphylococcus aureus in Palestine.
Community-acquired meticillin-resistant Staphylococcus aureus (CA-MRSA) is becoming an important public-health problem. This study attempted to investigate S. aureus and MRSA colonization in nasal swabs obtained from 843 patients without a history of hospitalization at the time of hospital admission and from 72 health-care workers chosen for comparison. Of the patients, S. aureus was detected i...
متن کاملMonitoring meticillin resistant Staphylococcus aureus and its spread in Copenhagen, Denmark, 2013, through routine whole genome sequencing.
Typing of meticillin resistant Staphylococcus aureus (MRSA) by whole genome sequencing (WGS) is performed routinely in Copenhagen since January 2013. We describe the relatedness, based on WGS data and epidemiological data, of 341 MRSA isolates. These comprised all MRSA (n = 300) identified in Copenhagen in the first five months of 2013. Moreover, because MRSA of staphylococcal protein A (spa)-t...
متن کاملPrediction of Staphylococcus aureus Antimicrobial Resistance by Whole-Genome Sequencing
Whole-genome sequencing (WGS) could potentially provide a single platform for extracting all the information required to predict an organism's phenotype. However, its ability to provide accurate predictions has not yet been demonstrated in large independent studies of specific organisms. In this study, we aimed to develop a genotypic prediction method for antimicrobial susceptibilities. The who...
